Purine-requiring and pyrimidine-requiring mutants of Saccharomyces cerevisiae.

چکیده
Haploid strains of S. cerevisiae were exposed to ultraviolet irradiation, and mutants were isolated by a modified layer plate procedure (Lederberg and Tatum, 1946), using a complete agar as the recovery layer. Six mutants were found which required adenine, two which required uracil. A seventh adenineless mutant was obtained through the courtesy of S. E. Reaume (Reaume and Tatum, 1949). Genetic analyses were conducted as previously described (Pomper and Burkholder, 1949; Pomper, 1951). Growth conditions were those of Schultz and Atkin (1947), volume and inoculum reduced 50 per cent, using the synthetic medium of Burkholder and Giles (1947). Growth is expressed as per cent absorption in a "lumetron" colorimeter, using a wire-glass filter.

Development of Saccharomyces cerevisiae as a model pathogen. A system for the genetic identification of gene products required for survival in the mammalian host environment.
Saccharomyces cerevisiae, a close relative of the pathogenic Candida species, is an emerging opportunistic pathogen. An isogenic series of S. cerevisiae strains, derived from a human clinical isolate, were used to examine the role of evolutionarily conserved pathways in fungal survival in a mouse host. As is the case for the corresponding Candida albicans and Cryptococcus neoformans mutants, S....
متن کاملCytokinin Utilization by Adenine-Requiring Mutants of the Yeast Saccharomyces cerevisiae.
By monitoring the growth of several adenine auxotrophs of the yeast Saccharomyces cerevisiae on cytokinin-supplemented media, we have demonstrated that this organism can utilize some of these derivatives as a source of adenine. Growth of a mutant lacking adenylosuccinate synthetase suggests that the conversion of cytokinins to adenine does not involve a hypoxanthine intermediate and may be cata...
متن کاملDegradation of Substituted Pyrimidine Nucleosides by Escherichia Coli and Bacillus Subtilis
It has been shown previously that hydroxyuridine competitively inhibits the growth of purine-requiring mutants of Escherichia coli (1). However, the inhibition is ultimately overcome, presumably because of the destruction of hydroxyuridine. It was of interest to det.ermine whether the inhibitor is, indeed, degraded, and, if so, what structural changes produce the loss of inhibitory activity. In...
